Structure Of Virulent Pathogen Revealed
Like high-profile politicians, pathogenic bacteria dispatch advance teams to make way for their arrival. But these bacterial agents don't just secure a safe passage, as a Secret Service detail might do. Rather they are teams of molecules that bacteria inject into cells they want to colonize, sent to hijack their hosts' biochemistry to serve their master's microbial needs. These molecules -- called virulence factors -- co-opt essential cell functions including the reproduction cycle and cell structure assembly, suppressing the cells' defenses against bacterial invasion and causing disease.

Effect of ghrelin on gonadotrophin secretion in women during the menstrual cycle
BACKGROUND

It has been suggested that ghrelin may affect reproduction in animals by decreasing pituitary LH secretion. The role of ghrelin on LH secretion in women has not been investigated. Our aim was to assess the effect of ghrelin administration on basal and GnRH-induced LH secretion during the menstrual cycle.

METHODS

Normally cycling women (n = 10) received on Day 3 of three consecutive cycles a single bolus i.v. of either ghrelin (1 µg/kg, cycle 1) or GnRH (100 µg, cycle 2) or GnRH plus ghrelin (cycle 3). In cycle 1, ghrelin was also injected in late follicular and mid-luteal phase of the cycle. Saline was injected in a preceding cycle (cycle 1, control). Blood samples were taken before drugs or saline injection (time 0) as well as at –15, 15, 30, 45, 60, 75, 90 and 120 min.

RESULTS

Plasma ghrelin levels increased rapidly, peaking at 15 or 30 min (P < 0.001), then decreased steadily, approaching pre-injection levels at 120 min. Serum FSH, LH, estradiol and progesterone levels remained unchanged. The stimulating effect of GnRH on LH and FSH secretion was unaffected by ghrelin injection. In contrast to saline, ghrelin stimulated a significant increase in growth hormone levels.

CONCLUSIONS

Under these experimental conditions, our results demonstrate for the first time the inability of a bolus of ghrelin to affect basal and GnRH-induced LH and FSH secretion. It is suggested that ghrelin does not play a major physiological role in gonadotrophin secretion in women.

Researchers have identified broad-spectrum aptamers. Aptamers are synthetic molecules that prevent the HIV virus from reproducing. In lab tests, aptamers known as RT5, RT6, RT47 and some variants of those were recently identified to be broad-spectrum, which would allow them to treat many subtypes of HIV-1. Field Evaluation of SD Bioline Rapid Malaria Diagnostic Test among Asymptomatic Malaria Infected Children in Port Harcourt, Nigeria
A rapid immunochromatographic test (SD Bioline malaria P.f/P.v) for the diagnosis of malaria was evaluated against thick blood smears in Port Harcourt, Nigeria, for the purpose of evaluating its efficacy for field work. Two hundred and forty children aged 1-8 years were surveyed in a cross sectional prospective study. For P. falciparum, the SD Bioline was 47% sensitive and 100% specific, with a positive predictive value (PPV) of 100% and a negative predictive value (NPV) of 83.2%. Efficiency of the test was 85.4%. The overall malaria prevalence in this study was 27.5%. P. falciparum accounted for 25.0%, while other species accounted for 2.75%. The cost of the SD Bioline test cassette (US $ 1.98 (230 Naira) per test at the time of study) was quite affordable. We concluded that the sensitivity of SD Bioline is low but with a threshold of 1000 parasites/microlitre among asymptomatic malaria infected children and affordable cost, this RDT (SD Bioline) could serve as a useful epidemiological tool in P. falciparum malaria endemic areas in the developing countries.

Coactivity of sulfide ingredients: a new perspective of the larvicidal activity of garlic essential oil against mosquitoes
BACKGROUND: The activity of several garlic oil ingredients against Culex pipiens L. larvae was studied. In addition to diallyl sulfide (DS) and diallyl disulfide (DDS), the garlic oils used included one essential oil (EO) and two semi-synthetic garlic essential oils (SSGEO1 and SSGEO2), which resulted from the enrichment of EO with DS and DDS standards respectively. The oils were compared with respect to their toxicity.RESULTS: The chemical composition of the tested oils was evaluated by means of gas chromatography-mass spectrometry. Experimental data from the tested samples revealed high toxicity. In detail, DDS (6.09 mg L-1) was the most active, followed by SSGEO2 (7.05 mg L-1) and EO (8.01 mg L-1), while SSGEO1 and DS were relatively inactive.CONCLUSION: The addition of DDS to EO did not change the toxic effect of the essential oil, whereas the presence of DS in excess produced an enriched essential oil with low toxicity. Furthermore, the antagonistic effect of DS and DDS against the other components of EO was shown by the application of two equations obtained from the literature. Their larvicidal performances were correlated, for the first time, with their detailed chemical composition. Data analysis strongly indicated the toxicity of the other EO sulfur ingredients. From the course home page: Course Description The lethal poison Ricin (best known as a weapon of bioterrorism), Diphtheria toxin (the causative agent of a highly contagious bacterial disease), and the widely used antibiotic tetracycline have one thing in common: They specifically target the cell's translational apparatus and disrupt protein synthesis. In this course, we will explore the mechanisms of action of toxins and antibiotics, their roles in everyday medicine, and the emergence and spread of drug resistance. We will also discuss the identification of new drug targets and how we can manipulate the protein synthesis machinery to provide powerful tools for protein engineering and potential new treatments for patients with devastating diseases, such as cystic fibrosis and muscular dystrophy. This course is one of many Advanced Undergraduate Seminars offered by the Biology Department at MIT. These seminars are tailored for students with an interest in using primary research literature to discuss and learn about current biological research in a highly interactive setting. Many instructors of the Advanced Undergraduate Seminars are postdoctoral scientists with a strong interest in teaching.
