Maternal depletion of NLRP5 blocks early embryogenesis in rhesus macaque monkeys (Macaca mulatta)
BACKGROUND

Genetic defects in the oocyte can lead to female infertility due to failures in early embryogenesis. NLRP5 (NLR family, Pyrin domain containing 5) is a maternal effect factor required for early embryo development beyond the 2-cell stage in mice. The NLRP5 gene is conserved in mammals. Applying RNA interference (RNAi) technique in the macaque oocyte, we studied the in vitro effect of NLRP5 in preimplantation embryo development in rhesus macaques.

METHODS

Macaque MII oocytes were collected and subjected to NLRP5 long double-stranded (ds) RNA microinjection, followed by intracytoplasmic sperm injection and in vitro embryo culture. NLRP5 mRNA and protein expression were examined, and the progress of preimplantation embryo development was monitored until the blastocyst stage.

RESULT

While 46.7 and 41.4% of embryos derived from uninjected and EGFP (Enhanced Green Fluorescence Protein) dsRNA-injected oocytes (control) reached the blastocyst stage within 10 days of culture, respectively, only 7.0% of the NLRP5 knockdown embryos developed to term. The majority of NLRP5 knockdown embryos were arrested between the 8-cell and the 16-cell stages.

CONCLUSIONS

The work provides the first evidence that long dsRNA-mediated RNAi can be applied to specific gene silencing in rhesus macaque oocytes, and demonstrates that maternal NLRP5 plays an important role in preimplantation embryogenesis in non-human primates.

Factors Affecting Delayed Diagnosis of Plasmodium vivax Malaria in the Republic of Korea
Estimation of the time from onset to diagnosis and analyses of characteristic factors was undertaken for vivax malaria cases diagnosed 1999-2003 and residing in non-malarious areas in Korea. Survival and multiple regression analyses revealed that those residing in provinces with major malarious areas and onset during the malaria season were diagnosed significantly earlier. It was considered that the identified factors reflect three important epidemiologic and ecological characteristics of vivax malaria in Korea: Spatially localized transmissions, clear seasonality in population dynamics of the vector and the partly prolonged incubation period.

5. Blogging to Enhance Learning Experiences Book Project

During the February Blogging to Enhance Learning Experiences seminar many participants expressed interest in writing a book together. Then things started to move very quickly! To date 34 members have joined the project by responding to a quick poll or enrolling in the book project space. We set up a wiki and began to carve out a table of contents and identify lead writers for chapters. We also have a forum for ongoing discussion about milestones, publishing options, copyright, and everything else that crops up with collaborative writing projects. You are welcome to join us! Choose your role: observer, editor, lead writer, contributer, reviewer, cheerleader... Any way you would like to contribute is appreciated as we write this book together!

Modelling binary mixtures of herbicides in populations resistant to one of the components: evaluation for resistance management
BACKGROUND: Herbicide mixtures are commonly proposed to delay the selection of herbicide resistance in susceptible populations (called the SM strategy). However, in practice, herbicide mixtures are often used when resistance to one of the two active ingredients has already been detected in the targeted population (called the RM strategy). It is doubtful whether such a practice can select against resistance, as the corresponding selection pressure is still exerted. As a consequence, the effect of mixtures on the evolution of an already detected resistance to one of the herbicides in the combination remains largely unexplored. In the present work, a simple model was developed to explore further the necessary and sufficient conditions under which a binary RM strategy might stabilise or even reduce resistance frequency.RESULTS: Covering the hypothetical largest range of parameters, 39% of 9000 random simulations attest that the RM strategy might theoretically reduce resistance frequency. When strong enough, high genetic cost of resistance, negative cross-resistance between the herbicides associated in the mixture and reduced selection differential between resistant and susceptible plants can counterbalance the resistance advantage to one of the two applied herbicides. However, the required conditions for an RM strategy to ensure resistance containment in natural conditions seldom overlap with experimental parameter estimates given in the literature.CONCLUSION: It is concluded that the sufficient conditions for an RM strategy to be effective would rarely be encountered. As a consequence, the strategy of formulating mixtures with herbicides for which resistance has already been detected should be avoided. Copyright © 2008 Society of Chemical Industry

Modelling The Effectiveness And Risks Of Vaccination Strategies To Control Classical Swine Fever Epidemics
In a recent update of the Dutch contingency plan for controlling outbreaks of Classical Swine Fever emergency vaccination is preferred to large-scale preemptive culling. Our modelling study evaluates the implications of this policy change. We find that vaccination in a ring of 2-km radius around a detected infection source is as effective as ring culling in a 1-km radius.

Plasmodium falciparum causes the most deadly form of malaria and accounts for over one million deaths annually. The malaria parasite is unable to salvage pyrimidines and relies on de novo biosynthesis for survival. Dihydroorotate dehydrogenase (DHOD), a mitochondrially localized flavoenzyme, catalyzes the rate-limiting step of this pathway and is therefore an attractive antimalarial chemotherapeutic target. Using a target-based high throughput screen, we have identified a series of potent, species-specific inhibitors of P. falciparum DHOD (pfDHOD) that are also efficacious against three cultured strains (3D7, HB3, and Dd2) of P. falciparum.
